Studies on the mechanism of superoxide release from human neutrophils stimulated with arachidonate.

چکیده
cis-Unsaturated fatty acids stimulate release of superoxide (O-2) by human neutrophils (Badwey, J. A., Curnutte, J. T., Robinson, J. M., Berde, C. B., Karnovsky, M. J., and Karnovsky, M. L. (1984) J. Biol. Chem. 259, 7870-7877). The rate of O-2 release due to arachidonate (105 +/- 24 S.D., nmol of O-2/min/10(7) cells) was comparable to optimal values obtained with other stimuli. Antagonists of calcium-binding proteins (i.e. phenothiazines, naphthalene sulfonamides) inhibited the release of O-2 in a fashion compatible with the involvement of calmodulin in these phenomena. Synthetic substrates for and an inhibitor of chymotrypsin-like proteases (e.g. N-benzoyl-L-tyrosine ethyl ester, L-1-tosylamido-2-phenylethyl chloromethyl ketone) also blocked O-2 release. Antagonists of calcium-binding proteins and of proteases were effective in this context with neutrophils stimulated with a variety of agents. The implications of these data for recent reports concerning the mechanism of action of cis-unsaturated fatty acids on phagocytes is discussed.

Effect of Neutrophils on Nitric Oxide Production from Stimulated Macrophages
Background: During the initial phase of an infection, there is an upregulation of inducible nitric oxide synthase in the macrophages for the production of nitric oxide. This is followed by the recruitment of polymorphonuclear leukocytes (neutrophils) which release arginase. Arginase competes with inducible nitric oxide synthase for a common substrate L-arginine. Objective: To investigate whethe...
متن کاملActivation and priming of neutrophil nicotinamide adenine dinucleotide phosphate oxidase and phospholipase A(2) are dissociated by inhibitors of the kinases p42(ERK2) and p38(SAPK) and by methyl arachidonyl fluorophosphonate, the dual inhibitor of cytosolic and calcium-independent phospholipase A(2).
Arachidonic acid (AA) generated by phospholipase A(2) (PLA(2)) is thought to be an essential cofactor for phagocyte nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity. Both enzymes are simultaneously primed by cytokines such as granulocyte-macrophage colony-stimulating factor (GM-CSF) and tumor necrosis factor-alpha (TNF-alpha). The possibility that either unprimed or cytokine...
متن کاملEvidence for participation of vicinal dithiols in the activation sequence of the respiratory burst of human neutrophils.
Phenylarsine oxide (PAO) specifically forms a stable ring complex with vicinal dithiols that can be reversed with 2,3-dimercaptopropanol (DMP). Pretreatment of human neutrophils with micromolar concentrations of PAO inhibited release of superoxide anion (O2-) stimulated by N-formyl-methionyl-leucyl-phenylalanine (FMLP) or phorbol 12-myristate 13-acetate (PMA); the inhibition was reversed with D...
متن کاملActivation of human neutrophil nicotinamide adenine dinucleotide phosphate, reduced (triphosphopyridine nucleotide, reduced) oxidase by arachidonic acid in a cell-free system.
Sonicates from unstimulated human neutrophils produce no measurable superoxide since the superoxide-generating enzyme, NADPH oxidase, is inactive in these preparations. Previous attempts to activate the oxidase in disrupted cells with conventional neutrophil stimuli have been unsuccessful. This report describes a cell-free system in which arachidonic acid (82 microM) was able to activate supero...
متن کاملSource of endogenous arachidonate and 5-lipoxygenase products in human neutrophils stimulated by bradykinin and A23187.
The lipoxygenase products of arachidonic acid (AA) metabolism, 5-hydroxyeicosatetraenoic acid (5-HETE) and leucotriene B4 (LTB4), are considered to have an important pathophysiological role in inflammatory bowel disease by stimulating the inflammatory response and by contributing to the diarrhoea. The present studies were designed to investigate the effect of the physiological stimulants bradyk...
